A highly efficient method for extracting peptides from a single mouse hypothalamus

Living organisms contain a variety of endogenous peptides that function as significant regulators many biological processes. Endogenous are typically analyzed using liquid chromatography–mass spectrometry (LC-MS). However, due to the low efficiency peptide extraction and abundance in single animal, LC-MS–based peptidomics studies have not facilitated an understanding individual differences tissue specificity abundance. In this study, we developed method followed by nano-flow LC-MS/MS analysis. This enabled highly efficient reproducible from sub-milligram quantities hypothalamus dissected animal. Diverse bioactive authentic were detected sample volume equivalent 135 ?g hypothalamus. may be useful for elucidating specificity, well facilitating discovery novel biomarkers developing therapeutics.
